Targeted Protein Degrader Building Blocks
PROTAC is a heterodimeric molecule, with one end of the molecule connected to a ligand that binds to a target protein, the other end connected to a ligand of an E3 ligase, and the middle connected by a suitable linker. The degradation of target proteins by PROTAC is achieved through the ubiquitin proteasome system (UPS). The general process is as follows: PROTAC molecules bind to target proteins (POI) and E3 ligases to form a ternary complex, labeling the target proteins with ubiquitination. The ubiquitinated proteins are recognized and degraded by the intracellular proteasome 26S.
